Born in London to Jewish parents, Lessore studied at the Slade School of Art. In 1931 she became secretary at the Beaux Arts Gallery, whose director was sculptor Frederick Lessore They married in 1934 and upon his death in 1951, she took over the directorship. Many of Lessore's artists gained distinguished reputations, including John Bratby, Frank Auerbach and Leon Kossoff. The work of Walter Sickert was regularly shown at the gallery until its closure in 1965.
