After graduating from St Anne's College, Oxford, Penston became a demonstrator in botany at the university in 1928. During her subsequent DPhil undertaken at Oxford, her research focused on the potassium nutrition of potatoes. In 1945 she became the first woman to be vice-principal of Wye College, University of London. She was appointed the principal of Bedford College in 1951 and remained there until she retired in 1964.
